Installing the hg19 Reference Genome Nirvana Annotation Database 1. Ensure that the DNA Amplicon Module version installed is version 1.1.0 or greater. This Nirvana Annotation Database is only compatible with version 1.1.0 or greater. 2. If present, Remove the following folder from your human hg19 genome: "\Homo_sapiens\UCSC\hg19\Annotation\Nirvana" (default reference genome repository: "C:\Illumina\Genomes\") (default reference genome repository MiSeq 2.x: "C:\Illumina\MiSeq Reporter\Genomes") 3. Download the "Nirvana.zip" file from the Local Run Manager Software Downloads Page. 4. Right click on the "Nirvana.zip" file and select "Extract All..." 5. In the "Extract Compressed (Zipped) Folders" dialog window, set the folder path where files will be extracted to the following location: "\Homo_sapiens\UCSC\hg19\Annotation\" 6. Once the extraction has completed, there should now be a \Homo_sapiens\UCSC\hg19\Annotation\Nirvana folder Within the Nirvana folder, the following folders should be present \Homo_sapiens\UCSC\hg19\Annotation\Nirvana\Cache\26. \Homo_sapiens\UCSC\hg19\Annotation\Nirvana\SupplementaryDatabase\44 \Homo_sapiens\UCSC\hg19\Annotation\Nirvana\Reference\5 (default reference genome repository: "C:\Illumina\Genomes\"). (default reference genome repository MiSeq 2.x: "C:\Illumina\MiSeq Reporter\Genomes") 7. Delete the "Nirvana.zip" file to save disk space.
